Pegula and Taylor Townsend competed in the main draw of the US Open doubles competition on August 30, 2011, after receiving a wildcard. Finally, they were defeated by the third-seeded combination of Vania King and Yaroslava Shvedova in the third round.

Pegula received a wildcard to the Indian Wells qualifying round in March 2012. She shocked top-ranked players Bojana Jovanovski and Paula Ormaechea to advance to the main draw, where she was defeated in three sets by Magdaléna Rybáriková.

Pegula qualified for the US Open and then played in her first major singles match there. She won over Margarita Gasparyan, Shuko Aoyama, and Melanie Oudin to get to the main draw, where she overcame Alison Van Uytvanck in the opening round.
